Sperm Preparation
Sperm cell preparation is an important step in the advanced treatment of infertility.
Sperm preparation distinguish functioning spermatozoa from those that are immotile, have poor shape, or can’t fertilize oocytes. It means that only the healthiest, strongest, and fastest sperm will be introduced to the egg in an IVF cycle.
Semen sample is analyzed to start the sperm preparation procedure. Our andrologist will determine the sperm count, the motility, and morphology.
Swim-Up Method
The Swim-up method is a sperm purification method used for in vitro sperm selection. This method is a reliable approach for separating motile sperm with acceptable morphology and higher chromatin integrity for your insemination.
The swim up is preferred if the semen sample has a normal number of sperms. A cell pellet or a liquid semen sample can be used for swim-up. A pre-washed sperm pellet collected by a soft spin is deposited in an overlying culture media in a conical tube in traditional swim-up. The most motile sperm is collected in the upper fraction, while sperm with little or no motility are collected in the bottom proportion.
The Process
- Semen is placed in a test tube; in some cases, it is previously washed and centrifuged.
- A culture medium is carefully placed on top of the semen. The medium is a hospitable environment for the sperm, and healthy sperm will swim into it. Slow and immotile sperm are is left behind, along with most debris in the semen.
- The test tube is let stand an hour or so; in some cases, it is placed at an angle, and/or in an incubator.
- In the standard procedure, the top layer is collected for use.
- The portion retained for use may be washed and centrifuged again.
